Test background

The fluorescent dye EMA (eosin-5-maleimide ) is used to label band 3 protein, which indirectly interacts with the spectrin-based cytoskeleton and membrane lipid bilayer in red blood cells.

Deficiency or abnormality of band 3 is seen in hereditary spherocytosis red cells, and has also been observed in cases of Southeast Asian ovalocytosis, congenital dyserythropoietic anaemia type II, and the cryohydrocytosis variants of stomatocytosis.
